Description
Cancer Antigen 125 (CA-125) also known as mucin 16 or MUC16 is a protein that in humans is encoded by the MUC16 gene. MUC16 is a member of the mucin family glycoproteins. It is more than twice as long as MUC1 and MUC4 and contains about 22,000 amino acids, making it the largest membrane-associated mucin. Serum cancer antigen-125 (CA-125) has long been used as a tumor marker for the preoperative diagnosis and monitoring of ovarian malignancy and may be a prognostic indicator for this disease. It is expressed by the coelomic and the Müllerian epithelium and can be elevated in various benign and malignant conditions, including pregnancy, menstruation, ovarian cancer, endometriosis, pelvic inflammatory disease, chronic renal failure, cirrhosis of liver, congestive heart failure, pleural effusion, colorectal and pancreatic cancer.

Intended Use
Cancer Antigen 125 Assay Kit is a chemiluminescent microparticle immunoassay (CMIA) for the quantitative determination of Cancer Antigen 125 (CA-125) in human serum and plasma. The assay kit is intended for in vitro diagnostic use.
Principle
The Cancer Antigen 125 assay is a quantitative sandwich immunoassay to determine the presence of CA125 in human serum and plasma using CMIA technology with flexible assay protocols.
- Sample, and paramagnetic anti-CA-125 coated microparticles are mixed, CA-125 present in the sample binds to anti-CA-125 coated microparticles, forming an antigen antibody complex.
- After incubation, a conjugate containing acridinium-labeled anti-CA-125 is added to the reaction mixture and binds to unoccupied binding sites of the anti-CA-125 coated microparticles.
- After further incubation and washing, Pre-Trigger and Trigger Solutions are added to the reaction mixture.
- The resulting chemiluminescent reaction is measured as relative light units (RLUs). There is a relationship between the amount of CA-125 in the sample and the RLUs detected by the optical system. Results are calculated automatically based on the previously established calibration curve.
Storage
The kit should be stored upright, not upside down or horizontally. Store at 2–8°C with a validity period of 12 months.
